Summary:

A Slovenian SME has developed a rapid cloning and gene manipulation platform for fast and efficient mobilization of large DNA fragments. The technology is based on one-step in vivo recombinational cloning. The technology allows for delivery of error-free DNA fragments and is used for cloning of large and complex gene clusters encoding bacterial secondary metabolites, or human immunoglobulins. Commercial agreement with technical assistance is sought by the company.

Details:

Conventional DNA engineering tools often present a major bottleneck for the highly complex demands of modern “synthetic biology” and metabolic engineering approaches. Upper size limits of DNA fragments in the classic cloning approaches are usually around several 10 kb, while direct DNA synthesis is limited both by the size of the synthesized fragments as well as by the risk of introducing errors from the originally published sequence. A Slovenian SME has developed a rapid one-step cloning and gene manipulation platform by combining the large accommodation capacity of artificial chromosome vectors (yeast artificial chromosome YAC), transformation-assisted recombination (TAR) cloning and recombining techniques for rapid manipulation of large gene clusters in S. cerevisiae, and their efficient transfer to the industrial heterologous hosts. This is achieved by constructing specific bi-functional “Large Pathway Cloning Vectors” (LPCVs), which enable selective in vivo cloning of desired large DNA fragments of interest in yeast. In the next step, the LPCV is isolated from yeast and transformed to E. coli, and finally delivered into the final heterologous host as a prominent secondary metabolite producer (e.g. a highly productive Actinomyces strain). The same technology can be efficiently used for the assembly of chemically synthesized DNA fragments opening the potential for efficiently generating synthetic DNA of up to several 100 kb in length.

When necessary for efficient expression, synthetic biology approaches are also being used by the Slovenian SME, including re-factoring or de novo synthesis of the entire gene sequence. The advanced gene tools platform allows for quick obtaining of the target sequence of interest for efficient manipulation and/or heterologous expression for production of the highly valuable bioactive metabolites from difficult-to-cultivate organisms.

The SME provides cloning technology as a stand-alone cloning service or can deliver the cloned DNA into a target microbial host strain. The SME can also further assess the host strain productivity in the scope of a full technology development project.

The company's research team numbers over 40 research scientists from diverse fields of biotechnology, microbiology, molecular biology, biochemistry and synthetic chemistry, with years of experience in the industrial strain improvement, bioprocess development, natural product biosynthesis and downstream/isolation procedures.

The company is looking for partners among biotechnology and/or pharmaceutical companies, academic/research institutions, or companies from other areas that have an interest in the technology. The company is interested in commercial agreements with technical assistance or technical cooperation.
